EDF - 30.1
EDF-30.1 is a metallic bioreactor with the working volume 23 l (total volume 33 l). The bioreactor is placed in an ergonomic stainless steel frame, which is folding, therefore, easily transportable. The bioreactor consists of the following main parts (see references in the picture):
- Frame
- Bioreactor vessel
- bioprocess controller BIO-3
- Peristaltic pumps unit
- Thermostat
- Control cabinet
- Air supply device
- Sterilisation/thermostat connection valves
- Water valve
The bioreactor is sterilisable in situ. For this purpose, the bioreactor jacket should be connected to the steam line with the help of switch valves. When sterilisation is accomplished, the bioreactor jacket is connected back to the thermostat using these switch valves.
At the compressed air input, there are an oil filter, a manometer and a pressure reduction actuator valve. Next are a stop valve, a rotameter, a microbiological filter and a relief valve.
On the frame's upper shelf, a peristaltic pumps unit and a bioprocess controller BIO-3 are placed. The peristaltic pumps unit (consisting of 3 pumps) ensures the pH control titrant liquid, the antifoam, and the substrate components' supply in the bioreactor, pumping these liquids from flasks (a shelf for placing the flasks is located in the back part of the bioreactor frame) through special needles in the bioreactor's upper lid. In the control cabinet is placed the frequency inverter, power switch, solid-state relay (for heater control).
The bioprocess controller BIO-3 controls:
- Temperature (the temperature measured in the bioreactor vessel is controlled by transmitting the control signals from the bioprocess controller to the thermostat heater or cooler. The circulation of the thermostated liquid through the bioreactor jacket is ensured with the help of a pump).
Thermostat consists of stainless steel heating vessel, in which is placed 1.5 kW heater. The cooling water in the heating vessel with the help of electromagnetic valve is injected. The contra pressure valve controls the direction of water flow. The circulation pump ensures the thermostating liquid flow through jacket of bioreactor vessel.
- pH (the supply of acid or alkali impulses in the fermentation liquid is ensured by transmitting the control signals to the peristaltic pump).
- pO2 (is regulated by changing the mixer's rotational speed. As an additional option, a combined pO2 regulation by changing the mixer's rotational speed and the amount of the supplied compressed air is possible).
- Foam (is regulated by transmitting the control signal to the antifoam peristaltic pump. As a measuring indicator, a foam sensor is used, which is installed in the reactor's upper lid and indicates electric conductivity).
- The mixer's rotational speed n (the stator poles of the electromagnetic drive are located outside the reactor vessel, and the magnetic rotor inside the vessel. The stator is directly connected to the frequency converter, which is controlled with the help of the bioprocess controller).
Altogether, the bioprocess controller BIO-3 has 16 analogous inputs (4-20 mA) and 16 digital inputs/outputs. Hence, there is a possibility to connect another 12 sources of signals of analogous values.
The bioreactor vessel consists of the following main parts:
- Upper lid
- Main part
- Stator
- Sampling device
The upper lid has holes that can be hermetically sealed. In these holes, the following devices can be installed:
- Needles - for the supply of substances capable of being titrated and metered;
- Manometer;
- Air condenser with a filter and an input valve;
- Temperature sensor;
- Chemostat tube;
- Foam sensor;
- Additional devices.
The main part is formed by the bioreactor vessel's parts with a jacket, the lower part and ports (in the lower part from the sides) for inserting Mettler-Toledo sterilisable pH and pO2 (D = 25 mm) electrodes.
Inside the Bioreactor vessel, there are:
- A fixed axis;
- A movable axis with a magnetic rotor and mixers;
- Baffles;
- Aerator.
In the lower part of the reactor, there is an electromagnetic stator, which embraces the reactor vessel from outside in its lower part. At the bioreactor vessel's bottom, a sterilisable screwed on sampling device (produced by the company "KEOFITT") is installed.
